Resistance to Meloidogyne arenaria in the peanut cultivar COAN is inherited as a single, dominant gene. The mechanism of resistance to M. arenaria in COAN was evaluated in three experiments. In the first experiment the number of second-stage juveniles (J2) of M. arenaria penetrating roots of the susceptible cultivar Florunner was higher than the number of J2 penetrating roots of the resistant peanut cultivar COAN (P < 0.05). In a second experiment it was determined that the root size and number of potential infection courts (root tips) were similar for the two peanut cultivars. The number of nematodes emigrating from roots of COAN after penetration was greater than emigrated from roots of Florunner (P < 0.05). Necrotic host tissue was rarely observed in roots of COAN infected with M. arenaria, suggesting that resistance to M. arenaria does not involve a necrotic, hypersensitive response. Most of the J2 observed in roots of COAN were restricted to the cortical tissue, with only 1 of 90 J2 observed being associated with the vascular cylinder, whereas in Florunner >70% of the J2 were associated with vascular tissues. Resistance in COAN may be due to constitutive factors in the roots.  相似文献

A mixed population of Meloidogyne arenaria race 1 and M. javanica race 3 is reported on peanut from a field in Levy County, Florida. Confirmation of M. javanica on peanut is based on esterase and malate dehydrogenase isozyme patterns resolved on polyacrylamide slab gels following electrophoresis, and perineal patterns. Up to 29% of 290 individual females collected from peanut roots in the field in autumn 2002 showed a typical esterase J3 phenotype for M. javanica. This is the third report of M. javanica infecting peanut in the United States.  相似文献

Changes in nutrient partitioning in stems and leaves of peanut (Arachis hypogaea L.) were examined after girdling of stems by Spissistilus festinus (Say) (Homoptera:Membracidae), the three-cornered alfalfa hopper. Under field conditions amino acids increased more than 12-fold in concentration in the 2 cm stem section above the girdle compared to below the girdle or control stems for newly formed girdles (less than 7–11 days). Asparagine and proline were the predominant amino acids, and increased 40- and 60-fold, respectively, above the girdle compared to below the girdle. There was no buildup of nutrient assimilates for the older girdled stems that had callused (and presumably recovered from feeding injury). A time course from 1–11 days was followed under greenhouse conditions, and girdled regions remained enriched in sugars for one day and amino acids for four days after girdle formation. After girdle formation most S. festinus responded to nutrient sinks by feeding within 5 mm above the girdle. This behavior was most pronounced one day after girdle formation, but persisted for seven days after girdle formation. During this period a new girdle was often formed within 10 mm above the original girdle. Under field conditions, stems and leaves derived from stems that had sustained an early or late season girdle generally had a reduced dry weight, total nitrogen content, and total carbon content; however, significant differences did not always occur. Thus, although temporary changes occurred in nutrient partitioning in girdled stems, there appears to be longer term negative growth effects on leaves and stems.  相似文献

Arachis pintoi is an alternative to forage production in the tropics. Its germplasm comprises more than 150 accessions, that could be used to improve it. Our objective was the isolation and characterization of microsatellite loci in A. pintoi to be used to molecular evaluation of this germplasm and of A. repens (section Caulorrhizae ). Seven loci were analyzed using five accessions of A. repens and 20 accessions of A. pintoi . The high variation found makes clear the high potential of this marker in genetic studies in these species. The developed markers showed total transferability to A. repens .  相似文献

花生种质资源是花生新品种选育和重要农艺性状遗传研究的基础材料。引进国外优异花生种质资源,并对其进行鉴定和评价对发掘优良花生种质、丰富我国花生资源遗传多样性以及合理利用种质资源进行遗传改良具有重要意义。本研究于2014-2015年连续2年在河北保定对104份引进的美国花生微核心种质资源纯化系进行了农艺性状考察和抗病性鉴定。鉴定结果表明,美国微核心种质纯化系多为匍匐型,主茎高变异范围为24.50~89.50 cm,侧枝长为39.37~99.23 cm,单株果数和单株果重分别为8.75~46.33个和8.49~29.54 g,百果重为80.76~216.72 g,单株粒数为18.25~58.00个,单株粒重为9.89~33.36 g,百仁重为25.52~74.18 g,出仁率为52.58%~76.08%。抗病性鉴定表明,部分美国微核心种质资源纯化系高抗褐斑病和网斑病,性状优良。该研究结果为花生新品种选育与遗传研究提供了优异材料和参考信息。  相似文献

对233份河南省地方花生资源进行了蛋白质含量、含油量、油酸和亚油酸含量的全面测定,并与省外和国外资源的相关性状进行了比较分析。在河南地方品种资源中,蛋白质含量中等,平均含油量和油酸含量相对较高,但缺乏蛋白质含量超过30%或含油量超过56%、油酸含量超过70%的突出材料。河南省目前高油品种选育有明显进展,育成了一批高油花生品种,但育成品种蛋白质含量普遍偏低。提出了充分利用现有地方品种资源,积极采用远缘杂交、诱变、分子标记辅助选择技术及现代基因工程技术创制优良种质,选育优质专用品种的育种策略。  相似文献

以花生主产区的棕壤和潮土为供试土壤,以种植面积最大的白沙1016、花育22和湛油27基因型花生(Arachis hypogaea)为供试作物,采用不添加Cd(对照)和添加Cd(1.5 mg·kg-1)处理进行盆栽试验,研究土壤类型和作物基因型对花生籽实Cd积累的影响.结果表明:两种土壤对照处理的3种基因型花生籽实Cd含量均低于国家食品安全标准,而Cd处理下均高于食品安全标准. 同种土壤Cd处理3种基因型花生籽实Cd含量显著高于对照,不同基因型间表现为湛油27>白沙1016>花育22,棕壤花生籽实Cd含量及总量均高于潮土. 对照处理3种基因型花生籽实Cd生物富集系数均大于1.0,Cd处理下多小于1.0,表明花生籽实对土壤中Cd的累积能力较强,土壤Cd含量进一步增加时,其生物富集能力降低.  相似文献

Hairy roots have the potential to produce a variety of valuable small and large molecules. The mist reactor is a gas phase bioreactor that has shown promise for low‐cost culture of hairy roots. Using a newer, disposable culture bag, mist reactor performance was studied with two species, Artemisia annua L. and Arachis hypogaea (peanut), at scales from 1 to 20 L. Both species of hairy roots when grown at 1 L in the mist reactor showed growth rates that surpassed that in shake flasks. From the information gleaned at 1 L, Arachis was scaled further to 4 and then 20 L. Misting duty cycle, culture medium flow rate, and timing of when flow rate was increased were varied. In a mist reactor increasing the misting cycle or increasing the medium flow rate are the two alternatives for increased delivery of liquid nutrients to the root bed. Longer misting cycles beyond 2–3 min were generally deemed detrimental to growth. On the other hand, increasing the medium flow rate to the sonic nozzle especially during the exponential phase of root growth (weeks 2–3) was the most important factor for increasing growth rates and biomass yields in the 20 L reactors. A. hypogaea growth in 1 L reactors was µ = 0.173 day?1 with biomass yield of 12.75 g DW L?1. This exceeded that in shake flasks at µ = 0.166 day?1 and 11.10 g DW L?1. Best growth rate and biomass yield at 20 L was µ = 0.147 and 7.77 g DW L?1, which was mainly achieved when medium flow rate delivery was increased. The mist deposition model was further evaluated using this newer reactor design and when the apparent thickness of roots (+hairs) was taken into account, the empirical data correlated with model predictions. Together these results establish the most important conditions to explore for future optimization of the mist bioreactor for culture of hairy roots. Biotechnol. Proximate and elemental composition of four peanut genotypes infected with groundnut rosette disease (GRD) was examined. Moisture and ash content generally decreased while fat and energy content increased in seeds from diseased plants. Protein and carbohydrate varied between seeds of diseased and healthy plants of the different varieties with no consistent pattern. Instrumental neutron activation analysis of 10 elements within leaves, stems and seeds showed elevated levels of K, Al and Cl in leaves, stems and seeds in at least three of the four varieties infected with GRD while Na was decreased in stems but increased in seeds. While significant differences were found, Mg, Mn, Ca and Zn did not show any consistent change with respect to plant part or genotype, between diseased and healthy plants. V and Fe were found at low levels in leaves and stems and not detected in seeds. This represents the first report on the effect of GRD on the nutritive quality of peanuts.  相似文献

The penetration of the intestinal mucosal wall is supposed to be critical for helminth parasite infestation, but has rarely been analyzed in detail. We here studied the establishment process of Schistocephalus solidus tapeworms in their second intermediate host, the three-spined stickleback, from oral uptake after experimental exposure, to passage through the gastro-intestinal tract and arrival in the fish body cavity. Using histological techniques, we found tapeworms to penetrate the intestine within 14-24 h, spending most of the time in the stomach lumen and only a very short period in the intestine. Unexpectedly, tapeworms lost their outer layer, together with the cercomer, in the intestine lumen rather than later during intestine wall penetration. Once exposed, the underlying tegument with microtriches might serve to facilitate migration of the parasite into the body cavity.  相似文献
